Duke Riley, artiste originaire de Boston, vit à Brooklyn, dans l'État de New York. Ancien tatoueur, il a vécu dans une pigeoniere pendant ses études à la Rhode Island School of Design au début des années 90, avant d'obtenir une maîtrise en beaux-arts à l'Institut Pratt. À la fin des années 90 et au début des années 2000, il a enseigné l'art dans des communautés défavorisées et des refuges pour victimes de violences domestiques à Boston et à New York, tout en poursuivant sa pratique artistique.
